Genetic Diversity of Chrysanthemum zawadskii Herb. and the Related Groups in Korea Using RAPDs


Abstract
Genetic diversity of Chrysanthemum zawadskii Herb. in Korea was examined using RAPD markers to complement conventional taxonomy of the plants. In addition, genetic relationship of this plants with other wild chrysanthemums in Korea and three chrysanthemum cultivars was examined using RAPD markers. C. zawadskii complex collected from 22 habitats in South Korea were divided into 1 species, 5 subspecies, and 2 varieties, including all taxa which were listed in previous several flora of Korea, though the number of taxa is debated among taxonomists. Genetic variation of C. zawadskii was studied using 8 screened primers with 72 RAPD bands. Genetic variation (relationship within species) of C. zawadskii was high enough to divide them into 3 large groups at subspecies level: such as C. zawadskii ssp. latilobum var. leiophyllum (Group II), C. zawadskii ssp. lucidum (Group I), and C. zawadskii ssp. acutilobum and 4 other subspecies (Group III). This grouping pattern seems to be influenced by ecological conditions. Group I1I could be divided into 3 subgroups at below subspecies level. Genetic diversity among genus Chrysanthemum and subtribe Asterinae, so-called wild chrysanthemum in Korea, was distinctly divided into above-mentioned two groups with a very low similarity. Genetic diversity among C. zawadskii, C. boreale, C. indlcum, and cultivated chrysanthemums within genus Chrysanthemum was separated into two subgroups. One group included C. zawadskii complex and the next group included 3 other species. Genotype of cultivated chrysanthemums seemed slightly closer to C. boreale and C. indicum than C. zawadskii. Morphologically similar species of Aster hayatae and Heteropappus hispidus, which belong to different genus each other, were found to possess close genetic relationship by RAPDs.
